When a person with Type A blood receives Type B blood in a transfusion, the ensuing reaction is potentially life threatening. IgM antibodies in the Type A individual cross-react with the red blood cells of the Type B donor, leading to massive clumping of antibodies and blood cells (agglutination). Our serologic tests take advantage of the same principle.
These tests rely on the fact that each individual antibody has two identical binding sites, meaning it will bind to two of its specific antigen (protein). In addition, IgM antibodies have very high affinity for each other and form a pentamer consisting of five IgM molecules and thus a total of ten antigen binding sites. From a practical standpoint, this means antibodies are very effective at linking antigens and antibodies together and causing massive clumping. For diagnostic testing, this means that the presence of a target protein or antibody can be detected by the presence or absence of agglutination. When an antibody in serum is the target to detect an infectious agent, its antigen will be included in our latex. Conversely, when a protein in serum is the target, its specific antibody will be present in our latex.
