Since its development in the 1960’s, ELISA (Enzyme-Linked Immunosorbent Assay) has revolutionized diagnostic testing, such as providing the first screening test for HIV.

By harnessing the high specificity of antibodies for their antigen, ELISA tests are able to directly detect specific components in serum. ELISA can detect either specific antigen (such as T3 for thyroid tests) or specific antibody (such as IgG/IgM for Rubella and other infectious diseases.) Although there are many variations, the tests generally work by binding antigen and antibody together on the base of a plate. A secondary antibody is added that binds to the primary antibody-antigen complex. The secondary antibody is linked to an enzyme that induces a color change when TMB solution is added, and the final color produced is directly proportional to the amount of antibody-antigen complex. This allows the concentration of the target antigen/antibody to be quantitatively determined with high accuracy using an ELISA absorbance reader.
